Ollivier Hyrien - Stochastic Modeling in Immunology

The goal of this lecture will be to introduce some of the most basic notions and terminology of the theory stochastic processes to immunologists. Those will include definition of the most popular stochastic processes (such as Markov chains…), their main characteristics (including the distribution function, the expectation, variance and covariance functions), and how to use and interpret stochastic processes. The lecture will first focus on Markov processes, which is the simplest type of stochastic processes. We will gradually reach more complex, but also more realistic models (non-Markov process).  Both continuous and discrete-time processes will be presented, and we shall discuss processes that model populations made up with one or several types of cells. Special attention will be given to branching processes, which find multiple applications in immunology and cell biology. We will also show how to fit such models to experimental data, with the main focus being on CFSE-labeling data. To make the lecture both attractive and understandable to experimentalists, we shall illustrate it through multiple examples coming from the fields of cell biology and immunology. For instance, we will demonstrate how stochastic processes be used to analyze cell division, cell differentiation and cell death. We will also draw a parallel between stochastic processes and discrete event modeling. Comparison with other modeling approach will be discussed.
